چکیده

Background and Objectives: Myeloproliferative neoplasms (MPNs) are Philadelphia negative disorders involving polycythemia vera (PV), essential thrombocythemia (ET), primary myelofibrosis (PMF). Although JAK2 mutation is almost involved, other several mutations linked to MPNs risk prognosis. TNFAIP3 genetic related cancers autoimmune diseases. Our study aimed demonstrate the effects of rs2230926_T/G & rs5029939_C/G SNVs gene on prognosis V617F positive MPNs.
Methods: Matched 2 groups in age, sex race were enrolled our research; 80 cases group 130 normal healthy controls with follow up for 3 years. Taqman assay probes involved real time polymerase chain reaction (PCR) utilized variants analysis.
Results: The rs2230926 rs5029939 modest linkage disequilibrium (LD) cases. observed frequencies G allele its genotypes both more prevalent patients than controls. bleeding symptoms presence splenomegaly existent heterozygous genotype combined respectively. overall survival (OS) was lower containing but progression free (PFS) affected only SNV.
Conclusion: revealed association increased incidence as well poor clinical course Egyptian ethnic.

Tumor Necrosis Factor Alpha -308 G/A Single Nucleotide Polymorphism and Risk of Sperm Abnormalities in Iranian Males

Objective Signaling molecules such as cytokines regulate spermatogenesis during the maturation of germ cells and apoptosis. Tumor necrosis factor-α (TNF-α) is one of the most-documented cytokines that supports spermatogenesis. We investigated the association of TNFα -308 Single Nucleotide Polymorphism with sperm abnormalities in Iranian population. MaterialsAndMethods This case control study in...

Leukemic blasts in transformed JAK2-V617F-positive myeloproliferative disorders are frequently negative for the JAK2-V617F mutation.

To study the role of the JAK2-V617F mutation in leukemic transformation, we examined 27 patients with myeloproliferative disorders (MPDs) who transformed to acute myeloid leukemia (AML). At MPD diagnosis, JAK2-V617F was detectable in 17 of 27 patients. Surprisingly, only 5 of 17 patients developed JAK2-V617F-positive AML, whereas 9 of 17 patients transformed to JAK2-V617F-negative AML. Microsat...

Pathogenetic role of JAK2 V617F mutation in chronic myeloproliferative disorders.

The molecular pathogenesis of chronic myeloproliferative disorders (MPDs) is poorly understood. The hematopoietic progenitor cells of patients with polycythemia vera (PV) or essential thrombocythemia (ET) are characterized by hypersensitivity to hematopoietic growth factors and formation of endogenous erythroid colonies. Recently, 4 groups reported almost simultaneously Janus kinase 2 (JAK2) V6...

JAK2 V617F: A Single Mutation in the Myeloproliferative Group of Disorders

The myeloproliferative disorders (MPD) are a group of haematological conditions where there is a primary disorder at the level of the multi-potent haematopoietic stem cell leading to increased production in one or more blood cell types. The three main disorders in the group are polycythaemia vera (PV), essential thrombocythaemia (ET) and idiopathic myelofibrosis (IMF). PV is characterised by an...

Evaluation of Tumor Necrosis Factor Alpha Polymorphism Frequencies in Endometriosis

Background The pro-inflammatory cytokine, tumor necrosis factor-alpha (TNF-α), is a pathogenic element for a number of disorders. Previous studies have reported that the -1031 T/C and -238 G/A polymorphisms in the promoter region of the TNF-α gene are important factors in reproductive-related disorders. One of the most common gynecological diseases of women during the reproductive years is endo...
